Of Mice and Men Literary Criticism Essay At the mention of the name, John Steinbeck, many associations can be made to the classic works produced by the man. However, he did not begin his life as a successful writer. Growing up in Salinas, California, Steinbeck lived the life of a common man, working to survive in the Land of Promise. He began to develop a taste for writing; however, he studied marine biology while he attended Stanford University. Without graduating with a degree, Steinbeck began working as a laborer and reporter for the American in New York City. After realizing that he was not meeting his goals, or at least coming close to them, Steinbeck moved back to California where he continued to work in various odd jobs while he pursuing his career as a professional writer. In the early 1930s Steinbeck met Edwards Ricketts, a marine biologist who later became a major influence on his works. Steinbeck enjoyed listening to Rickettss views on the interdependence of life. Steinbeck became intrigued by these concepts and began applying these themes to all of his developing works. His first two publications were undeveloped and deemed failures, but with the production of Of Mice and Men, Steinbeck was introduced to the world of literary acceptance. Of Mice and Men is considered an American classic that is taught by almost every high school and featured on most must read booklists. The reason for such appraise can be found in the themes lying within the book. Steinbeck establishes the concepts of mans destiny by developing his characters, the relationships they share, and their interactions and using them to reinforce underlying themes. The relationship between Lennie and George opens many types of interpretation for critics of the novel. Some wonder whether George cares for Lennie and a person, or maybe George only feels obligated to watch over him. Is it a matter of Georges sincerity, or does Steinbeck want to reinforce the idea that humans would naturally want to be alone? Of Mice and Men tells the story of two men who depend on each other to survive. Their relationship resembles one that would be found between two brothers where one, George, is looking out for the other, Lennie. This relationship can be illustrated through out the novel. When the boss asked Lennie what he was capable of doing, George replied for him with, He can do anything you tell himhe can do anything. Just give him a try (22). George noticed that Lennie might no be able to give the boss the right answer so he decided to help out and take matters into his own hands. He does this because he feels that Lennie is his responsibilityand he continues to do this throughout the book. Lisca takes notice of the relationship and says that it could be seen in Georges sacrifices and devotion to Lennie (348). Devotiong is definitely the word to use because George was devoted to Lennie. George had a choice: whether he should take on Lennies responsibilities or not. Steinbeck uses George and Lennies companionship to set them apart from the rest of the people. Not only do they have a dream, but they do have each other. Before the two arrive at the ranch, the two discuss exactly what it is that sets them apart from the rest of the rest of the ranchers when George says, We got a futureIf them other guys gets in jail they can rot for all anybody gives a damn. But not us (14). George is letting Lennie know that they are above the others, that they have a plan to get out of the rut they are in. French also finds this element in Steinbecks work. When George shoots Lennie, he is not destroying only the shared dreambut also destroying the thing that makes him different and reducing himself to the status of an ordinary guy (349). It is evident that this dream made the two stand out among the rest. Steinbeck wants to emphasize the idea of loneliness by creating characters and depicting them as being alone in the world, despite the fact that they are surrounded by other people. With the growing number of people supporting the legalization of marijuana is it time to pass laws and make recreational use legal, or do all faults outweigh the good? The debate on whether marijuana should be legalized is currently a controversial topic. Many people have expressed their views on the topic, supporting their reasons for whether they think marijuana should be legalized. For some states it was easier to decide if the pros of legalizing marijuana outweighed the cons; such as Colorado and Washington. These states have already legalized the recreational use of marijuana. Marijuana is a fast-growing bushy plant with dense sticky flowers. It has been a part of our American culture for hundreds of years. It has been referred to as a poison, drug and narcotic. It is believed that marijuana was introduced by the Spanish in 1545 to Chile. In America the plant was first grown in Virginia and Massachusetts in the sixteen hundreds. It became a major commercial crop that was grown along with tobacco and was a source of fiber for fabric, rope and cloth. Later it was used to treat many health problems. During the 19th century many medical articles were written discussing marijuana’s value in treating conditions. The plant was considered legal from the 1600’s until the 19th century. Marijuana became popular in the mid-19th century and was in most drugstores through the beginning of the 20th century. It was around this time that recreational use of marijuana was becoming acknowledged. Marijuana has been illegal in the United States since t he Marijuana Tax Act of 1937. This is a federal law that made the possession or transfer of marijuana illegal throughout the United States. This law was the precursor to the crimina... ...ression, motivational syndrome, and even death. The brain damage has been shown to cause memory loss and difficulty in problem solving. In males, it will lower testosterone levels, lower sperm count, and may cause impotence. In females, marijuana may have some effects on pregnancy and can also cause birth defects. Legalization of marijuana could eventually lead to the legalization of harder drugs. Slowly progressing till it becomes possible for harder drugs to gain acceptance. Drugs like heroin, cocaine, and amphetamines, which many view now as unacceptable could eventually be up for legalization. Both sides give legitimate reasons whether it was for or against the legalization and should be taken into consideration. In the end when all sides of the argument are considered, it is simply a matter of opinion on whether you think marijuana should be legalized.

Alice Walker and James Baldwin both use their literature characters to bring social problems to light. The following will compare and contrast how Baldwin and Walker's characters use this connection as a means to sort through their â€Å"despair†. Every Day use by Alice Walker is a story narrated by a mother that has a very appropriate title.This title refers to the character, Maggie, whose sister says will use the heirloom quilts made by their grandmother every day when they should be hung Like she would do with such priceless Items. Maggie sorrowfully says It's okay; her sister can have the quilts. The mother sees such a selfless daughter, who in fact feels worthless in comparison with her sister and most likely several people, considering her po or self-image. She abruptly rips the quilts from her more dominant daughters' hands, Dee, and hands them over to Maggie.The way Alice Walker uses these characters to portray race, lass, gender and society Is shrewdly done. The mother who is possibly a farmer when reading the description: â€Å"l am a big boned woman with man working hands† can represent class (1334 Charter). She Is a low class citizen who lives In a small home that Dee, her daughter, doesn't approve of. Her daughter, who goes to college and has broken out of the lower class ranks, can also represent class. In addition, they represent race. They are of African American ethnicity and living during the oppressed times, according to â€Å"whoever can imagine me looking a white man in the ye? Spoken by the mother (1334 Charter). Dee does mention the fact that her sister Maggie can make something of herself â€Å"It's really a new day for us but form the way you and AMA still live, you'd never know It† (134 0 Charter). This sentence can represent the gender and race issues within those times. Dee was a black female a double burden because both were discriminated against at this time which brings us to society. Although this references the views of society, Walker does not use the mentioning of outside characters to portray society like Baldwin does.She manages this with only the main characters of the story. Society is for the reader to assume when peering Into the torn relationship of the sisters†¦ TLS relationship has been molded by outside factors that frown on the weak (Maggie has scars and uneducated) and glorify the beautiful (Dee is attractive and educated). Baldwin, on the other hand, makes great use of society in his story Sonny Blues, which is also suitably titled. This story is narrated by a brother who is asked to take care of his younger brother Sonny by his passing mother.Sadly, Sonny is a heroin addict, hence the title, and always In trouble. His brother doesn't wan t to deal with It, for obvious reasons: one being that Is too hurtful to go through but In the end he was able to find the strength after the loss of his little girl to make amends with Sonny and take him under his wing. The characters that represent society in this story are many. They are all inner city poor residents. There is the druggy that tells the narrator about Sonny â€Å"l see you got the papers so you know about it? † he asks him of Sonny (59 Charters).The barmaid dancing on page 60 and the children who are playing in the oppressed residents off poor and uneducated area. Very similar to Walker, he uses the main characters to reveal issues of class, race and gender. The narrator, who can represent class, has broken away from poverty by receiving a college degree and becoming a high school math teacher. The brother, can be made to represent race of the inner city. He has not broken free from the black oppressed lifestyle that his neighborhood has to offer. The fact that Baldwin uses males as main characters eaves the reader to assume gender roles.It is a silent assumption that men are able to be strong and should not need the help of others. This type of thinking tears the brothers apart when the older brother assumes his little brother can â€Å"get life on his own† keeping him from taking care of his little brother like his mother asked of him. â€Å"l sensed myself in the presence of something I really didn't know how to handle† says the narrator of his role in Sonny's life (67 Charters). Both authors use the characters to expose social issues within their times.

In today’s journalistic world it is sometimes difficult to decipher between news broadcasts rendered as credible information as opposed to merely entertainment. It is safe to say that the media does have a responsibility in reporting the news truthfully and with honesty. As we begin to look more closely on the role of the News Media, local and national, we will find many interesting facets of the broadcasts to be analyzed. It is also expected that the media deliver news that is detailed, informative and credible.Unfortunately the content of the information sometimes broadcasts has other insignificant focuses such as, news regarding celebrities, entertainment (especially here in Los Angeles so close to Hollywood) and other tragic but unnecessary news. The content seems at times unimportant, weak, and falsely dramatic rather than a factual honest and necessary report of important occurrences local, national and international. At times the news seems more like a dramatic soap ope ra rather than news. Competion and monetary gain seems to drive the media to serve information that can be sometimes manipulated or sometimes just simply inappropriate.Another facet of the news media is the status of female anchors and reporters on network and local newscasts, which will be examined in this paper. It is important to note that the media has been known to be a field influenced by gender, racial and ethnic biases. These important topics will also be examined. It seems that the News Media is not very particular in choosing quality journalism these days. News broadcasters are more influenced by events that will cause an immediate impact and sensational effect on the viewers than actual, credible and significant news. It is common for news broadcasters to begin the news  with such broadcasts covering such events as a â€Å"Wild Pursuit† or freeway car chase.Or, other such glamorized topics may include the arrest of famous actors or musicians whom have committed crimes. The O. J. Simpson case was a perfect example of such media frenzy or, the famous pop musician George Michaels being arrested for masturbating in the Beverly Hills Park restroom. How can the media choose to cover such insignificant events when the local areas, the nation or the international community has so many other important social, economical, political and humanitarian problems and events that are hardly addressed.Also, when we pursue to analyze the credibility of broadcasters, how are we sure that the information given to us is factual? Are we to believe everything that CNN reports is untainted and free of excessive nonfactual events and information? It would be justified to question the source of reported events and news stories. There has been so many times in which different views and different versions of the same story have been reported by varying news channels. Therefore it is sometimes difficult to decipher between fact or fiction and credible information or merely entertainment.It is also interesting to note that the media tries to maintain an air of social and political correctness when it comes to choosing its journalists although certain biases still due exists in the journalistic field. It is only in recent times that the news media has decided to hire more ethnically diverse journalists. Also in a field that had bee n mostly dominated by men, in recent times we have seen the hiring of more female anchors, reporters and journalists. Sometimes broadcasters use attractive journalists to gain the interest of the viewers.One of the most important TV news personalities of the last 40 years has been the famous Barbara Walters. She is one of the first women to gain acceptability and credibility from television news viewers. Also Christian Amanpour remains the first female TV broadcaster in our history who commands the same kind of respect as the long line of father figures that started with Murrow and continues with Rather, Brokaw and Jennings. Women journalists such as Walters and Amanpour have gained respect and stardom through many years of hard work and struggle through journalism.Nowadays, historic news stories have become the journalistic fast track to celebrity. And this happens so routinely that the search for the new media stars automatically built into coverage of the events themselves. Again this proves how today’s news media functions more as entertainment than credible responsible journalism.Some may say that it does not take any journalism skill to read a news script, and most stations want an appealing person, so why not have a pretty person reading a copy of the news? Personally I believe that this is unfair to other journalists male or female who have pursued a career studying and experiencing modern journalism. It is unfair to hire â€Å"sexy† or â€Å"sexual –attractive† journalists to report the news when the most important factor is the conten t and truthfulness of the news itself. It may be appropriate to flaunt ones sexuality in the fictional soap opera or fictional TV show arena, but not in the modern journalistic forum.Gender and sexuality should not be a factor. It would be hopeful to expect the News Media to cover more events with greater importance and significance to all people rather than to use journalism as a form of entertainment for the masses. Why complicate our language lives with the choice? Is the universe malicious? According to one study, the suffix -ic is preferred over the variant -ical by a ratio of 8 to 1. Curiously, however, when -log precedes the suffix, the ratio is reversed. (In another example of this phenomenon, called potentiation, -ness is much more common than -ity except when the suffix is preceded by able.) But that doesn’t answer my questions. For the most part, the choice seems to be personal or institutional preference, because there’s usually no distinction no logic, for example, to selection of -logic or -logical. For example, the style guide of the American Academy of Neurology prefers the shorter form, but in other contexts, neurological prevails. One researcher points out that, as you might have guessed, -ic (from the Greek suffix -ikos) was the original suffix; -ical, formed by adding the French suffix -al, came later. For the most part, usage organically caused a divergence, so that, for example, a historic occasion is memorable, whereas a historical occasion is one that merely occurred. For another example, economic refers to economics, while economical is used more generally to refer to the quality of economy. In this case, as with some others, the former can mean the same thing as the latter but seldom does. Comic and comical, and geometric and geometrical, are two of the many other sister terms with both (occasionally) identical and (usually) distinctly different meanings. Sometimes, one form predominates for obvious reasons (fanatical, for example, developed in favor of fanatic because the original form came to be applied as a noun), but in other cases, the variation for reasons seldom clear triumphs (botanical versus botanic, for example.) So, which form should you use in a given context? The dictionary is helpful for most -ic/-ical debates, but the -logic/-logical (and -logous!) issue is an outlier. ABSTRACT Aquaculture is the farming of aquatic organisms such as fish, crustaceans, molluscs and aquatic plants. Disease emergence is a concern in wild fisheries due to environmental pressures, the direct impact of human activities and the risk of pathogens spread from aquaculture. Common viruses are Taura syndrome virus (TSV), white spot syndrome virus (WSSV), and the necrotizing hepato pancreatitis bacterium (NHP-B). The White Spot Syndrome Virus is the most economically devastating viral pathogen to global shrimp aquaculture production and has been proposed to be capable of infecting all decapod crustaceans. WSSV is an enveloped ellipsoid virus, which belongs to the genus Whispovirus of the family Nimaviridae. VP28 is one of its major envelope proteins, and plays a crucial role in viral infection. In this study, the proteins of the infected crab were purified using SDS page and then Western Blotting was performed to extract that particular protein. INTRODUCTION White spot syndrome virus (WSSV) is one of the major shrimp pathogen that causes a high mortality rate of 90-100% within 3-10 days of infection (Lightner, 1996). Natural WSSV infections have been found in captured and cultured specimens of the mud crab. WSSV is an enveloped ellipsoid virus, which belongs to the genus Whispovirus of the family Nimaviridae. In addition, WSSV can also infect a wide range of hosts including both decapod and non-decapod animals with more than hundred species described to date. So far, the genome from three different WSSV isolates has been sequenced. Sequence analysis showed that WSSV contains approximately 500 putative open reading frames (ORFs), most of which have no homology with any known genes or proteins in public databases. Till now, more than 50 structural and non-structural proteins were identified. Tools such as polymerase chain reaction (PCR) and nested PCR, originally developed by Lo etal., have been widely used and recommended by the Office of International Epizootics (OIE) to be used as standard diagnostic methods for the detection of WSSV. Despite their excellence in specificity and sensitivity, these methods were not suited in some circumstances due to their complications, the requirement of thermal cycler, time-consuming, and labor-intensive. Moreover, the classical agarose gel electrophoresis with ethidium bromide staining, following the visualization under the ultraviolet (UV) transilluminator required to analyze the result of PCR products. Western blotting identifies with specific antibodies proteins that have been separated from one another according to their size by gel electrophoresis. The blot is a membrane of nitrocellulose or PVDF (polyvinylidene fluoride). The gel is placed next to the membrane and application of an electrical current induces the proteins in the gel to move to the membrane where they adhere. The membrane is then a replica of the gel’s protein pattern, and is subsequently stained with an antibody. Therefore, these features could be limited their applications, particularly in the resource-limited areas and non-laboratory environments such as at the pond or station sites. 2. MATERIALS AND METHODS 2.1. Tissue homogenate preparation: Gills, muscle, hepato pancreas and Head soft tissue from the crab infected with WSSV were homogenized in 1:10 suspension with NTE Buffer. It was then freeze and thaw for three times then centrifuged at 5000 rpm for 5min. The supernatant were collected separately in a tube and stored at -20Â °C. This supernatant was used for protein analysis. 2.2. Protein estimation: Lowry’s method was performed for the estimation of protein in order to know its concentrations (Lowry et al., 1951). 2% sodium carbonate in 0.1N sodium hydroxide (Reagent A) 0.5% copper sulphate in 1% potassium sodium tartrate (Reagent B) Alkaline copper solution: Mixed 50 ml of solution A and 1 ml of B prior to use (reagent C) Folin Ciocalteau reagent (Reagent D) Protein standard stock: 50 mg of bovine serum albumin (fraction V) was weighed and dissolved in distilled water and made up to 50 ml (1 mg/ml) Working standard- diluted 1ml of stock solution to 5 ml with distilled water. (200 Â µg of protein/ml). 2.3. SDS-PAGE: SDS-PAGE is currently used to determine the molecular masses, and also to investigate the subunit composition and the domain structure of proteins. TAE Buffer (1X) was poured in tank which maintain the temperature and pH of the gel. Gel was prepared and protein sample is mixed with bromophenol blue which act as a tracking dye. Then it was loaded into the well and electric current of 50V is applied into the tank which is later on increase to 100V when the dye contacts the resolving gel. When the dye reach the end of the resolving gel it was taken out and stain with the help of Coomassie Brilliant Blue R-250 (methanol-50 ml, distilled water 40 ml, glacial acetic acid 10 ml) for 1 hour and de-stain the gel by immersing in the de-staining solution (65 ml of distilled water, methanol 10 ml and 25 ml of glacial acetic acid). 2.4. Western blotting: Transferring of the protein into a membrane The membrane can be either nitrocellulose or PVDF. PVDF requires activation with methanol for one minute and is then rinsed with transfer buffer before preparing the stack. After stacking, the electro transfer is carried out for 90 minutes at a constant current of 150 mA set by maintaining the voltage at maximum. Transfer to the membrane and checked using Ponceau Red staining before the blocking step. (PVDF cannot be stained). After ponceau staining, wash the nitrocellulose membrane with TBST for 1 min at room temperature. Blocking and Antibody incubation Block the membrane for 1 hour at room temperature in blocking buffer (10 % milk in TBST). After blocking, incubate the membrane in primary antibody (1:1000 dilutions) in 2.5% milk in TBST overnight at 4Â °C. Wash the membrane for three washes using TBST for 5 minutes each. Incubate the membrane with secondary antibody (1:2000 dilutions) in 2.5% milk in TBST at room temperature for 1 hour. Wash the membrane in three washes of TBST, 5 minutes each. Add ECL reagent (Solution A and B in 1:1 ratio) on to the membrane and spread it thoroughly on to it using a pipette. Remove the excess reagent and place the membrane on to the x-ray cassette and expose to x ray film in the dark room. An initial 10 sec exposure should indicate the proper exposure time because due to the kinetics of the detection reaction, signal is most intense immediately following incubation and declines over the following 2 hrs. 3. RESULTS AND DISCUSSION 3.1. SDS-PAGE: Fig.1 SDS-PAGE which shows different bands under white light. Lane 1- Gill; Lane 2- HST; Lane 3- Muscle; Lane 4-Hepatopancreas. The polypeptides of purified virus were resolved by SDS-PAGE using discontinuous buffer system. Western blotting Viral polypeptides separated by SDS-PAGE were electrophoretically transferred to nitrocellulose membrane (0.22 ÃŽ ¼m) in transfer buffer (192 mM glycine, 25 mM Tris and 20% methanol, pH 8.3), at 0.8 mA/cm2for 1 h. it was later on view by using film to visualize the relative abundance of proteins Discussion By performing SDS-PAGE, (fig.1) we can determined the different molecular weight of polypeptide. The difference in the number of the polypeptide may be attributing due to the difference in concentration of the resolving and other physical conditions. The western blot analysis of proteins of semi-purified white spot syndrome virus separated on 12.5% gel (fig.2) showed different number of immunogenic proteins based on its molecular weight. Out of these, those proteins, which stained more intensely, were considered as major immunogenic proteins. However, further studies are needed to establish its immunogenic nature and feasibility for its use as vaccine. Because electrophoretic separation of proteins is always carried out under denaturing conditions, the problem of solublization, aggregation and co-precipitation of target proteins with adventitious proteins are eliminated. America's Flawed Constitution Right from the beginning of it’s creation the constitution of the United States has been a shaky document. The very basis for it being there was in fact illegal. The story of American politics starts with the Declaration of Independence. This document was brilliantly written by Thomas Jefferson and compacted all of the great ideas of enlightenment into one short easy to read paper. The declaration stated all of the ideals the new American nation would strive for. A constitution was needed as a way in which to fulfill those goals. The articles of confederacy were created as that constitution. However, they were weak, because no state wanted to give away any of their powers, and so the articles eventually failed. That is when the modern day constitution was starting to form. The Articles of Confederacy stated that in order to change any part of the document all thirteen states must agree to the change. Therefor a meeting was called so that they could amend the failing articles. However, representatives from two of the states did not show up. Even though not all states were represented the meeting started and the first vote was to totally throw away the Articles of Confederacy. The constitution wasn’t formed yet and it was already a flawed document. Because not all states were represented when the articles required it, the constitution was an illegal document. The delegates working on the constitution new that they needed a stronger document, because the articles proved too weak, but it still needed to please all of the states. This was impossible. So what ended up happening was the new ducocument became more and more vague. The only way to create a document that would pass was to make a document which didn’t really solve any problems but make each state believe that there problems would be fixed. This was accomplished by making it so that it was too vague to offend anybody but you could read into it. This made for a document that would be seriously flawed because people would be able to read into it too much. It could not work. The Constitution of the United States of America was too vague to work. The way the constitution was written it gave power to four parts: the congress, the executive branch, the judicial branch, and the states. Because it was so vague it did not really define which powers went where (with a few exceptions).

Character Education and Me Essay

After reviewing all of the materials for the course, I am convinced that character education is something that must have a presence in schools. Students of all grade levels can benefit from learning how to become civilized members of society. I agree with Lickona (1992) as far as the family being the first and foremost teacher of morals. As adults, we continue to reflect on our morals and virtues with hope of continually improving ourselves, especially when we become parents. I as well as many other new parents ran out to get a parenting manual to assist me in raising my children the best I could. Character education should be incorporated into the lessons daily in context. As the School Family video showed us, it should not be done through bribing, rewarding and punishing students without a lesson involved. After the school adopted the Conscious Discipline program, it appears that they have students use their words to talk out their issues with one another. I feel this has a better impact on the students in the long run. They will be better able to handle conflict in the future because of the lessons their teachers have helped them to understand. Kohn (1997) explained the side of character education with which most teachers are familiar. He explains, much like the Fern Creek video states, that students are most frequently drilled and rewarded through the character education programs. I do not necessarily agree that the values should be taught out of context. As a language teacher, I can tell you first hand that rote memorization does not show understanding of a concept. In addition, any person who has raised a preschooler can tell you that one of the first questions a child asks is â€Å"Why? † Children do not learn from simply being corrected or told what to do. They learn when they can explain why something is the way it is. Good character is completely developed by knowing ones morals and wanting to abide by the virtues by which we all live. If more parents were sensitive to the information that Lickona discussed in the Raising Good Children video, schools would have an easier time with character education. I agree that parents and schools need to form a partnership. Parents typically do not line their children up and drill right or wrong. They teach their children virtues in context. If my son steals his brother’s toys and a fight breaks out, I am not likely to say â€Å"that’s not right! † and end the conversation there. Young children are more likely to stop the bad behavior if they know why it was wrong or hurtful. I would explain to my son, â€Å"It hurts your brother’s feelings when you take his toys. Do you like your feelings hurt? Please ask him next time. † Incorporating the â€Å"do unto others† ideal does not make an impact unless the child can relate directly to the situation. I have noticed the times I had forgotten the explanation of why to stop; my son would repeat the misbehavior a short time later. Many of the virtues I have learned have come from my mother and my grandmother, since they were around me the most as a child. Through gentle reminders from these women I learned a set of standards to live by, my virtues. By knowing this information and using the good moral judgment that my family taught me, I have developed my character. Some children come to our classrooms without the opportunity to learn the way I did. Therefore, I need to be sensitive to their needs and help them to focus on how to change their behaviors. By treating students fairly and with compassion on a consistent basis, hopefully I can encourage students to intrinsically want to have good character.
